To select a normal or non-reactive antibody control, pick an antibody from the same host as the primary antibody.

For antibodies made against recombinant proteins, we provide a reference sequence. The exact epitope to which the antibody binds is not known, as we do not conduct epitope mapping. Many ADMEbio antibodies are made against short synthetic peptides derived from the target protein. ADMEbio uses the proprietary algorithm Epinetix_TM developed by our scientists to select these peptides, and hence they are not available for distribution; however, we will be happy to test for homology against the same target from other species and report that information to you.

Immunoglobulins extracted from serum consists of many molecules with slight variations in sequence which cause them to bind to different proteins and/or different binding sites on the same protein. Individual IgG molecules are categorized by isotype based on similarity of amino acid sequences. Monoclonal antibodies are secreted from immortalized cell lines called hybridomas which have been derived from B-cells of immunized animals. Each monoclonal antibody is of a single isotype because they are derived from the identical clone.

Polyclonal IgG consists of the entire spectrum of antibodies produced by all clones generated in response to an immunization. A polyclonal IgG therefore does not have a single isotype but consists of the mixture of isotypes found in the serum of the immunized host.

The observed band size may be different from the predicted band size for two main reasons:

Migration may not be exclusively due to protein size – Migration rate is subject to relative charge variation, and/or posttranslational modifications that cause the protein to migrate through the gel differently than expected based solely on the molecular weight of the protein.

The apparent molecular weight of the protein may be different than expected - because the molecular weight of the protein is actually different than that calculated from amino acid sequence. This can occur due to cleavage or degradation of the protein, splicing or the formation of protein multimers.

Cross reactivity information is supplied from in-house testing data or verification of 100% sequence homology. Predicted cross reactivity is based on targets with a great than 80% sequence homology of the peptide immunogen. Antibodies with whole protein immunogens have an increased likelihood of reacting with similar proteins from other species, or with family members from the same species.
